Project: Pre-clinical genotype-phenotype predictors of Alzheimer's disease and other dementia

The main aim is to establish genotype-phenotype matching in incipient Alzheimer's disease (AD) and Lewy-body diseases (Lewy) and Parkinson’s disease (PD). Common factors causing dementia development in these diseases, and common genetic variants each conferring a small risk, may be involved. Using large GWAS studies, large cohorts of established disease cases and large pre-dementia longitudinal studies combined with advanced imaging and neurochemistry we will identify the polygenetic risk factors for neurodegenerative dementias and investigate how these new "pre-morbid (ie genetic) markers" together with "pre-dementia and pre-clinical phenotypic markers" improve prediction of disease development, and likely form latent etio-pathogenic pre-dementia classes relevant for prevention strategies. Reduced variance for prediction of clinical disease progression from an earlier to a more advanced stage (e.g., from MCI to dementia) will enable improved prognostic guidance of individual patients, and is essential for treatment studies, in terms of increased power from the studies. This will increase the likelihood of detecting significant effects when relevant, and reduce the costs for development of new preventive agents.
